Hello,
Here is a test scenario that i created to illustrate the problem.
There are two identical submarines: SUB A and SUB B. Both subs are at periscope depth (-66ft).
SUB A is unassigned and its periscope and esm sensor are working correctly.
SUB B is assigned to ASuW mission and it's basically "blind". Neither periscope nor esm sensor seems to be working.
So it looks like subs assigned to missions are "blind" when on periscope depth.

Setting the SUB B Patrol Depth to -65 feet seems to solve the issue, the contact is detected by both active radar and eyeball (i.e. periscope). You have radars set to active on the Sub B patrol, so Sub B fires up it's active radar system.

Also if you manually set the sub to "Periscope Depth" with the Unit Orders function, things work as intended. Perhaps setting the depth to exactly -66 ft on a patrol mission does not give the signal to raise scope and start radiating.

Okay looked at this one...

You set the depth to exactly -66ft which is 20.11m. The simulator core uses the metric system, and periscope depth is -20m. As such, the sub is evaluated as being submerged.

If you go to Game Options, turn off 'Altitude in feet', change it to -20m, and then turn 'Altitude in feet' back on, it will display -66ft but the altitude is actually -20m rather than -20.11m. And it should work correctly.
