It IS mentioned in manual - Major Regimes are protectionists and don't allow free trade between them initially. You need to play diplomacy stratagem to unlock trade between you and another major. AFAIK you can use filters when selecting your leaders. And when selecting a zone - you can simply click on it on mini-map :) Yes some do have filters but send spies for example has only a list of zones in a kind of 'random' order, now clicking on the region has the same issue I pointed out above with the Strat Map where the 'click' is way below center of where I click so ends up being 'guess work' but i'm not sure if you have a similar issue or if it may be connected to monitor size, map size or such.
