How do you get a replacement Glen onto a sub which has lost its plane?

Park the sub somewhere where there is more that 20,000 in supply and set it to accept replacements.

Sometimes it can take a while if the unit has zero planes- even if you have a pilot or two. So if a small unit has zero planes I upgrade then downgrade back down again and you get your plane quick- but you have to pay twice the supply. It would be too expensive for a medium or large unit- but these don't normally go to zero planes.

I got confused because if the sub is missing its plane then you can't get to the aircraft screen. I checked all my other subs which still have aircraft to make sure accept replacements is on and now hope that's the case with the sub missing its plane. Not sure how to pull off that upgrade trick without being able to get to the aircraft screen. How can that be done if the sub has no planes onboard?

The planes are not directly attached to subs.
They are attached to air groups which in turn is attached to subs.

Even if no planes, you should be able to click on the air group.
Open up TF screen, click on the sub and you should see the air group (if one) in the information screen to click on.

Not ALL subs cary Glens, only a few. Click on the sub in question and see if a Glen airgroup is attached to the sub, if so, you can see the size of the airgroup. If the size is 0, it has lost the entire airgroup and will be needing a replacement Glen. Get that in a port with 20K od supply.........

The sub in question can have a Glen onboard, checked that. So I now understand that I must move my sub to a 20K+ supply base to have the chance to get another plane. Thank you all for the information.

But now I have an "out of curiosity" question related to my first question. If I go to the TF screen of that sub, it shows Planes: 0 and it will not allow you to go to the airgroup screen. So, if for some reason I had Allow Replacements turned off before I lost my Glen, how would the sub ever get a plane back? Or does that not matter at this point since there is not an airgroup associated with the sub at this time?

Click on the blue airplane on the top row of your screen. The "planes at sea". At the bottom, click the turn on replacements for all airgroups option (you probably want the vast majority of your airgroups at sea to have replacements anyway. If not, you can turn off various types, such as fighters for example if you dont want to modify their current settings). Until you get to know the game, its probably best if you leave replacements on. Later, you'll tweek it to your own taste.
