What are these new comprehensive difficulty settings and how do they work or function in the game? How many levels of difficulty are there?

You can see the settings, and a little overview here in case you missed it:

Thanks Spacecadet that was very helpful and of course got me antsy to buy the game. I'm so tired of playing lame games with lame ai and the pirates sound like they are a pita but how are they and the other empires later in the game when you breakout? Do they maintain some kind of power and relentless attacks on you?

I think you will find Shadows (especially if you play in the Age of Shadows) much more of a challenge than Legends.

Pirates do have their positive side (Smuggling, Attack Missions, etc.), but playing as an Empire, I generally prefer to not to deal too much with them unless I have.

- They extort from you
- They try to sneak in and get control of your planets
- They're usually competing for the same resources as you, and can actually choke off your supplies to certain resources

These are just the more obvious ones [;)]

If you start in the age of Shadows the pirates are a pita for a long time, also the default starting conditions for a game in the Age of Shadows has plentiful pirates so they will be a pain for extra long. It's a good challenge though if you are looking for it, really something you will struggle to deal with on harder difficulty settings. The normal age pirates aren't much worst than the old days, just different in how they behave and interact with your empire.
You can of course tweak the starting game settings of amount of pirates and amount of conflict, and of course make it so pirates don't respawn.
Pirates are also a lot harder to eradicate because their ability to build new space ports for themselves.

Setting the difficulty up really does provide a challenge even to very experienced players, I currently play not on the hardest setting and struggle for most of the game to keep up with other empires.
