to make a rail transport a unit needs to be in a town or city. You need resources to do a rail transport. The city hex of the destination has to be empty and all rail hexes inbetween have to be undamaged.
Damaged rails are marked like in the picture below.
If all is set up to meet the conditions, city hexes that can be reached will be highlighted.
